Moderna stock is where the mrna stock searches landed on Tuesday, and the number that turned the day into a story is the pullback in a 545 percent year-to-date rally. William Blair cut the biotech to Market Perform from Outperform on Monday, Oct 26, citing valuation. In Tuesday trading shares were about 3 percent below Monday's $198.85 close near the open, dipped as low as $179.53 intraday, and hovered near $195 at midday (Yahoo Finance daily prices, retrieved Oct 27).

The move is the market's reaction to a simple claim: the reward is already banked. Investing.com reported on Oct 27 that William Blair analyst Myles Minter called the current level aggressive after the stock's roughly 250 percent surge since positive Phase 3 news in August. The firm kept its oncology thesis but argued the shares had priced in the upside. Citi, which retains a Sell rating with an $80 price target, was already on the other side, and Brookline Capital Markets trimmed its long-term earnings estimate in the same wave of cautious notes.

None of it was cheap to get here. The rally traces to interim data from the Phase 3 INTerpath-001 trial of intismeran, Moderna's personalized neoantigen vaccine, given with pembrolizumab in adjuvant melanoma. Presented at the ESMO Congress and relayed by Investing.com on Oct 27, the results convinced William Blair the combination supports a potential new standard of care if Merck and Moderna can launch it commercially. Minter's research note, dated Oct 26, still frames the ceiling narrowly: the firm models $5.4 billion in peak melanoma sales, shared 50:50 with Merck, against a Keytruda franchise he expects to reach $35 billion in 2028 before genericization.

The gap explains the valuation test. William Blair's sum of the parts analysis implies a 70 to 75 percent probability of success across additional tumor types, according to Investing.com on Oct 27, and the total net present value sits just above the current market capitalization. With Moderna valued near $79.4 billion, that thin margin of safety is what the downgrade is really about. The stock has already given back 17.9 percent over the past week, per Investing.com analyst data.

What happens next is a binary readout calendar. Investors are waiting on the Phase 2 INTerpath-004 trial in renal cell carcinoma, which Investing.com said on Oct 27 is expected to deliver results by late 2026 or 2027 and could determine whether intismeran matches checkpoint inhibitor commercial potential or stays a melanoma franchise. Phase 2 programs in renal and bladder cancer are expected in 2027, Benzinga reported the same day. In the nearer term, Moderna has scheduled a manufacturing facility tour in Marlborough, Massachusetts for Nov 12, a marker for how the company plans to scale a personalized product.

For holders, the bull case is that melanoma proof of concept is real and the failure risk in additional tumors is what a 70 to 75 percent implied success rate is meant to capture. The bear case is arithmetic: when a stock that ran 545 percent trades on 70 percent odds priced in, even good news can disappoint. The dates to watch are the renal readout, the Marlborough tour, and whatever the next analyst note does with those two.
